玉米低聚肽改善肝纤维化作用机制研究及其在饮料中的应用Study on the Mechanism of Corn Oligopeptides Improving Liver Fibrosis and Its Application in Liver Protection Drink

摘要(Abstract):
为研究玉米低聚肽对肝纤维化的影响,以昆明小鼠为研究对象,灌胃CCl4建立肝纤维化模型,灌胃不同剂量的玉米低聚肽溶液给予治疗,同时给予1mg/kg剂量的秋水仙碱作为阳性对照。通过苏木素-伊红(hematoxylineosin,HE)染色和Masson三色染色评估肝纤维化小鼠的肝损伤和胶原沉积;通过酶联免疫吸附测定法(enzyme linked immunosorbent assay,ELISA)、反转录聚合酶链反应(reverse transcription PCR,RT-PCR)和蛋白免疫印迹法(western blotting,WB)研究玉米低聚肽对肝纤维化的改善机制。HE染色和Masson三色染色结果表明玉米低聚肽能够使受损的肝组织结构恢复正常,且减少了肝组织胶原沉积。此外,ELISA、RT-PCR和WB结果表明玉米低聚肽通过降低促炎因子水平和减少细胞外基质的产生从而改善肝纤维化。上述结果表明玉米低聚肽具有护肝的效果,且基于玉米低聚肽的护肝效果进一步研究一种以玉米低聚肽为核心原料的有益于肝脏的饮料配方,为其作为新型益肝饮料的开发提供了理论支撑。
